Edward Jenner created the first vaccine in 1796 using cowpox.

He noticed milkmaids who caught cowpox never got smallpox. He deliberately infected a boy with cowpox, then exposed him to smallpox — the boy didn't get sick. The word 'vaccine' comes from 'vacca' (Latin for cow).
